PABA/NO抗人鼻咽癌细胞株CNE细胞增殖和迁移的作用

摘要 目的探讨PABA/NO对体外培养人鼻咽癌细胞株CNE增殖和迁移的影响及其机制。方法将体外培养的处于对数生长期的CNE细胞随机分为PABA/NO组及对照组,PABA/NO组分别予0-6~10-10mol/L浓度的PABA/NO进行干预,对照组不干预。48 h后采用MTT比色法和细胞计数法检测CNE细胞增殖情况;Transwell小室法检测CNE细胞迁移情况;硝酸酶还原法检测细胞培养液中一氧化氮(NO)浓度;RT-PCR法检测细胞血管内皮生长因子(VEGF)mRNA表达。结果与对照组比较,PABA/NO干预后CNE细胞增殖和迁移均受到抑制(P<0.05),且该效应在10-6~10-8mol/L范围内具有较好的浓度依赖关系,细胞培养液中的NO浓度明显提高,VEGFmRNA表达下调。结论 PABA/NO能抑制CNE增殖和迁移,其机制可能通过提高NO浓度,抑制肿瘤血管生成,从而发挥其抗肿瘤作用。 Objective To investigate the effects and mechanisms of PABA/NO on the proliferation and migration of nasopharyngeal carcinoma cell line CNE.Methods CNE cell in logarithmic phase cultured in vitro was divided into PABA/NO group and control group,the PABA/NO group was treated with PABA/NO at 10-6~10-10 mol/L,the control group was not treated.The MTT assay and cell counting method were used to examine the proliferation of cell;Transwell chamber assay was performed to determine migration of cells;enzyme reduction of nitrate was used to detect the concentration of NO in culture supernatants of CNE;the VEGF mRNA was detected by real time RT-PCR.Results Compared with the control group,the cell proliferation and migration were both inhibited(P〈0.05),and in PABA/NO dose-dependent manner at the concentration of 10-6~10-9 mol/L,and the NO concentration increased significantly in culture media,the expression of VEGF mRNA decreased significantly.Conclusion PABA/NO has marked inhibitory effect on proliferation and migration of CNE cells,the mechanism maybe related to up-regulate the concentration of NO,downregulate the expression of VEGF,and so to play anti-tumor role.
