黄芩茎叶总黄酮对Aβ_(25-35)致大鼠学习记忆损伤及海马抗氧化酶活性的影响 被引量:8

摘要 目的:探讨黄芩茎叶总黄酮(scutellaria baicalensis stem-leaf total flavonoid,SSTF)对大鼠海马注射Aβ25-35致大鼠学习记忆损伤及海马抗氧化酶活性的影响。方法:给大鼠灌胃(ig)SSTF 100,50,25 mg.kg-1剂量,采用双侧海马注射Aβ25-35制作大鼠痴呆模型,Morris水迷宫实验测定大鼠学习、记忆能力;HE染色观察大鼠海马CA1区神经元形态;检测海马组织中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活性及丙二醛(malondialdehyde,MDA)含量。结果:模型组大鼠寻找平台潜伏期较对照组大鼠明显延长(P<0.01),2 min内在平台象限所游路程与总路程的比值明显低于对照组(P<0.01),SSTF 100,50 mg.kg-1剂量组及维生素E组的大鼠寻找平台潜伏期较模型组明显缩短,所游路程比值明显高于模型组;模型组大鼠海马CA1区神经元损伤明显,SSTF100,50 mg.kg-1剂量组及维生素E组大鼠海马神经元损伤较模型组明显减轻;SSTF100 mg.kg-1剂量组海马组织中SOD、GSH-Px活性较模型组明显升高,MDA含量明显减少。结论:黄芩茎叶总黄酮能减轻大鼠海马注射Aβ25-35引起的海马神经元损伤,改善学习记忆能力,其机制可能与提高组织中抗氧化酶活性有关。 Objective: To explore the effect of scutellaria baicalensis stem-leaf total flavonoid (SSTF) on learning and memory damage induced by Aβ25-35 and on the activity of anti-oxidative en- zyme. Methods: Rats were administered SSTF (100,50,25 mg.kg^-1.ig). The AD model was made by bilateral injection of Aβ25-35 in rat hippocampus. Morris water maze was used to evaluate the learning and memory ability of the rats. HE stain was used to observe morphology of the neuron of hippocam- pus CA1 region. The activity of superoxide dismutase (SOD),glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in rat hippocampus were measured. Results: Compared with thatof the control group, the latency to find the platform in model group was significantly increased, and the ratio of path in platform quadrant to the whole path was significantly decreased. Compared with that of the model rats, the latency to find the platformin animals with SSTF 100, 50 mg-kg-~ and vitamin E (Vit E) groups were significantly decreased, the ratio of path in platform quadrant to the whole path was significantly increased. The CA1 region neuron of the model rats showed severe damage. Compared with of the model rats, the neuron of SSTF and Vit E groups showed a decreased damage. Activities of SOD, GSH-Px in SSTF and Vit E group were significantly increased compared with that in the model rats. Content of MDA in SSTF and Vit E groups were significantly decreased compared with that in the model rats. Conclusion: SSTF can attenuate neuron damage and learning impairment induced by Aβ, and the mechanism may be related to the increased activity of anti-oxidative enzyme.
