鲤鱼汤对阿霉素肾病大鼠CD2相关蛋白及肾病蛋白的影响

Effects of Cyprinus Carpio Decoction on the expression of nephrin and CD2AP in adriamycin nephropathy rats

目的:观察阿霉素肾病大鼠肾小球足细胞CD2相关蛋白(CD2-associated protein,CD2AP)和肾病蛋白(nephrin)的表达及鲤鱼汤对其表达的影响,进而探讨鲤鱼汤治疗肾病综合征的作用机制。方法:70只Wistar大鼠随机分为5组:正常对照组(N组)、模型组(M组)、福辛普利组(F组)、鲤鱼汤低剂量组(YD组)和鲤鱼汤高剂量组(YG组)。M组、F组、YD组和YG组大鼠采用尾静脉注射阿霉素6.5mg/kg复制阿霉素肾病模型。第3周开始进行干预,YG组和YD组分别予以高、低剂量鲤鱼汤(2.25、1.13g/mL)灌胃7周;F组予以0.09mg/mL福辛普利灌胃;N组和M组予以等量自来水灌胃。采用免疫组化方法检测各组大鼠肾脏中CD2AP、nephrin的表达,并计算其阳性表达的平均光密度值(AOD)。结果:与模型组相比,鲤鱼汤高剂量和鲤鱼汤低剂量组大鼠血清总蛋白水平、血清白蛋白水平等均有一定升高,尤其鲤鱼汤高剂量组差异均有统计学意义(P<0.05);各治疗组肾小球中CD2AP与nephrin的表达较N组均降低,但与M组比较,都有不同程度的增加(P<0.05)。结论:鲤鱼汤可不同程度地增强阿霉素肾病大鼠肾小球足细胞CD2AP和nephrin的表达,改善阿霉素肾病大鼠肾小球的损伤,进而起到治疗作用。

Objective： By observing the expression of CD2AP and Nephrin on adriarnycin nephritic rat kidney, and the effects of Cyprinus carpio decoction on it, to explore the mechanism of cyprinus carpio decoction on nephritic syndrome. Methods： 70 rats were randomly divided into 5 groups： normal control group（N group）, model group（M group）, Fosinopil group（F group）, low Cyprinus carpio decoction group（YD group） and high Cyprinus carpio decoction group（YG group）. The nephropathy model was induced by injecting adriamycin 6.5g/kg from candas vein. The levels of CD2AP and nephrin expression were detected by immunohistochemistry method, and the average optical density （AOD） was calculated. Results： Compared with N group, the expression of CD2AP and nephrin decreased in treatment groups （P〈0.05）, especially in the M group; compared with the M group, the expression of CD2AP and nephrin increased in F group, YD group and YG group （P〈0.05）. Conclusion： The results of this study show that cyprinus carpio decoction play a importment role by improving the expression of CD2AP and nephrin on adriamycin nephropathy rats, and alleviating injury of glomerular filtration membrane in adriamycin nephropathy rats.
