配伍对桃核承气汤中苦杏仁苷代谢分解的影响研究 被引量:6

Influence of compatibility difference on the content change of amarogentin in Taohe Chengqi Decoction

摘要 目的:研究配伍对桃核承气汤中苦杏仁苷体液代谢分解的影响。方法:采用HPLC法测定苦杏仁苷的含量,通过L8(27)正交实验设计,考察桃仁不同配伍煎液经人工胃肠液体外代谢前后苦杏仁苷的含量变化,并进一步运用统计软件(SPSS 13.0)对结果进行分析。结果:①方中各药材对人工胃液处理前后苦杏仁苷的影响次序为:大黄>芒硝>桂枝>甘草,其中大黄、芒硝的影响具有统计学意义(P<0.05),而桂枝、甘草有影响但无显著性差异;②方中各药材对人工肠液处理前后苦杏仁苷的影响次序为:大黄>桂枝>芒硝>甘草,其中大黄的影响具有统计学意义(P<0.05),而其余药材有影响但无显著性差异。结论:大黄和芒硝可能是影响桃核承气汤中苦杏仁苷代谢分解的主要因素,具有促进桃仁消化和苦杏仁苷分解的作用。本研究可为桃核承气汤的减毒机制研究提供理论基础,同时为同类中药复方的配伍机制研究提供参考。 Objective: To find the reasonable factors that affect the amarogentin contents in Taohe Chengqi Decoctions (TCD). Methods: Eight different sample solutions were analyzed through L8(27) orthogonal design and statistics analysis (SPSS 13.0). Content changes of amarogentin in these solutions were determined by HPLC. Results: (1) In the experiments of artificial gastric juice, it was Rheum palmatum L.and Nitrum that significantly impactedthe content change of amarogentin (P〈0.05), while for Radix Glycyrrhizae and Cortex Cinnamomi, the difference of those change were insignificant;2) In the experiments of artificial intestinal juice, it was Rheum palmatum L. that significantly impacted the content change of amarogentin (P〈0.05), while for other tablets, the difference of them were insignificant. Conclusion: The decomposition of amarogentin in TCD may be mainly impacted by Rheum palmatum L. and Nitrum. This study can be helpful to develop the mechanism research of TCD compatibility.
