摘要
本实验改良了传统方法,自创PF膜和浮膜培养法,首次整装培养人血管内皮细胞成功,经高锰酸钾固定剂固定,消化细胞内蛋白质,保留脂质膜性系统,应用透射电镜进行整体观察。在此基础上,用氢过氧化枯烯处理,造成整装内皮细胞线粒体,内质网的脂质过氧化损伤模型。本实验进一步证实脂质过氧化对内皮细胞生物膜的损伤,并报告了内质网和线粒体损伤的整体结构变化。
The whole-mount culture human endothelial cells is firstly succeeded withimproved traditional method of the whole-mount culture cells and culture methodof PF membrane and floating. membrane creaed in this paper.Treated with cumenehydroperoxide, then fixated with potassium permanganate fixative, proteinskeleton system was melted and digested but lipid membrane system was fixatedand retained in the cells. The membrane system eould be observed under TEM.The Injury degree of mitochondria and endoplasmie reticulum in endothelialcells induced by the method mentioned above was of differencnce in this experi-ment. The higher cumere hydroperoxide concentration is, the severer lipidmembrane system injury is. The injury of endothelial cells is considered atrigger in atherogenesis, thus lipid peroxide might play an important role inreducing atherosclerosis.
