摘要
目的探讨大蒜素对卵巢癌耐顺铂细胞株SKOV-3/DDP凋亡的影响及机制。方法将SKOV-3/DDP细胞随机分为四组,大蒜素组加入40μg/mL的大蒜素,顺铂组加入40μg/mL的顺铂,联合组加入大蒜素和顺铂各40μg/mL,对照组不干预。各组分别培养24、48 h。采用MTT法检测各组细胞增殖抑制率;RT-PCR法测定Bax、Bcl-2mRNA表达;Western blot法测定Bax、Bcl-2蛋白表达。结果顺铂组、大蒜素组、联合组各时点(24 h、48 h)增殖抑制率明显高于对照组,联合组明显高于大蒜素组、顺铂组,P均<0.05。顺铂组、大蒜素组、联合组各时点Bax mR-NA和蛋白水平明显高于对照组,大蒜素组、联合组Bcl-2 mRNA和蛋白水平明显低于对照组,P均<0.05;顺铂组48 h Bcl-2蛋白明显高于对照组,P<0.05;联合组各时点Bax mRNA和蛋白水平明显高于、Bcl-2 mRNA和蛋白水平明显低于对照组,P均<0.05。结论大蒜素诱导SKOV-3/DDP凋亡作用优于顺铂,两者联合具有协同作用,其机制可能为上调Bax表达和下调Bcl-2表达有关。
Objective To investigate the effect and mechanism of Allicin on the proliferation of cisplatin-resistant o- varian cancer cell line of SKOV-3/DDP. Methods The SKOV-3/DDP cells were divided into four groups randomly, the Alliein group was treated by 40 μg/mL Alliein, the cieisplatin group was intervened by 40 μg/mL cicisplatin, the eonbine group was treated by 40 μg/mL A11icin and eieisplatin, the control group was treated by nutrient fluid. All the groups were cultured for 24 hours and 48 hours. The cellular growth inhibition rate was calculated by MTI'. RT-PCR was used to detect the expression of Bax and Bcl-2 mRNA ; Western blot was used to detect the protein level of Bax and Bcl-2. Results The growth inhibition rate of the Cisplatin group, allicin group, combination group at each time point (24 h, 48 h) was signifi- cantly higher than that in the control group, which in the combination group was significantly higher than that in the cisplatin group and the alliein group, all P 〈 0.05. The Bax mRNA and protein levels of Cisplatin group, alliein group and combined group at each time point were significantly higher than those in the control group, which in the Alliein group were significantly lower than those in the control group ( all P 〈 0.05 ) ; the expression of Bel-2 protein in cisplatin group at 48 h was significantly higher than that in the control group ( P 〈 0.05 ) ; the Bax mRNA and protein levels in the combined group at each time point were signifieandy higher than those in the control group, but the Bcl-2 mRNA and protein levels were significantly lower than those in the control group, all P 〈 0.05. Conclusions The SKOV-3/DDP apoptosis-inducing effects of allicin is superior to cisplatin, teombination of the two drugs has a synergistic effect, its mechanism may be related to the upregulation of Bax expression and downregulation of Bcl-2 expression.
