RNA干扰Tax基因表达对人嗜T淋巴细胞病毒Ⅰ型相关性脊髓病/热带痉挛性瘫痪患者细胞凋亡的影响

摘要 目的探讨RNA干扰Tax基因表达对人嗜T淋巴细胞病毒Ⅰ型相关性脊髓病/热带痉挛性瘫痪(HAM/TSP)患者细胞凋亡的影响。方法运用RNAi技术特异性干扰Tax基因的表达,流式细胞技术和westernblot法鉴定Tax蛋白的表达情况,并运用流式细胞技术对干扰后的T淋巴细胞(TLC)凋亡情况及凋亡相关因子Fas、Bcl-2、肿瘤坏死因子α(TNF-α)进行检测。结果重组质粒psiTax/U6转染T细胞(9.84±3.32)%后,Tax蛋白的表达较未处理组(47.45±14.52)%和pSilencerl.O/U6转染组(45.98±10.66)%明显降低(t=9.51,14.93,均P〈0.01);westernblot法显示重组质粒psiTax/U6转染TLC后,Tax表达较pSilencerl.0/U6转染组明显降低;RNAi干扰组的凋亡率(21.55±1.37)%较未处理组(47.80±3.36)%显著降低(t=24.17,P〈0.01);RNAi干扰组Fas,TNF—α显著降低(t=17.83,16.85,均P〈0.01),而Bcl-2显著升高(t=5.21,P〈0.01)。结论运用RNA技术干扰Tax基因抑制Tax基因表达,可以有效抑制TLC细胞的凋亡。凋亡相关因子Fas,TNF—αandBcl-2在此过程中发挥重要作用。
