摘要
目的:优化大孔树脂分离纯化巴豆中木兰花碱的工艺条件。方法:采用HPLC法测定木兰花碱,筛选AB-8,HPD400,ADS-17,NKA-9,D101树脂对木兰花碱的吸附和解吸附能力,并优化分离纯化条件。结果:优选出D101树脂,上样液浓度为生药1.5g/mL,吸附流速1.5BV/h,药材量:树脂量为6∶1;分离纯化条件为:先用4BV水冲洗除去不吸附杂质,再用3BV 8%乙醇除去大极性干扰杂质,最后用4BV 23%乙醇冲洗收集目标滤液。结论:该方法简便易行,适用于巴豆中木兰花碱的精制与纯化。
Objective: To study the optimal optimally purified process of magnoflorine extracted from Croton tiglium with macroporous res- ins. Methods: The HPLC was used to analyze the content of magnoflorine,the adsorption and desorption performances of AB-8, HPD400, ADS-17,NKA- 9,D101 macroporous resins were compared and then optimize purification conditions. Results: D101 resin was selected because it gave the most effective of separation. The optimum process for the purification was as follows: the concentration of sample solution was 1. 5 g/mL; adsorption flow rate was 1.5 BV/h; crude Medicinal materials weight than reisn weight for six. 4 BV water 3 BV 8 % ethanol and 4 BV 23 % ethanol were used as etuant of Croton tiglium respectively. Portions of 23 % ethanol were collected. Conclusion: This method is simple and feasible with good effect of enrichment in magnoflorine from Croton tiglium.
