阔叶十大功劳叶柄愈伤组织诱导及试管苗培养的研究 被引量:3

摘要 以大连市引种的阔叶十大功劳嫩叶柄为外植体,采用组织培养方法研究其快繁技术,结果表明:愈伤组织诱导的初代和继代理想培养基是MS+KT 0.6 mg·L-1+2,4-D 2.5 mg·L-1;不定芽分化的理想培养基是1/4MS+ZT0.4 mg·L-1+NAA 0.1 mg·L-1;不定芽生根的理想培养基是1/3MS+IAA0.5mg·L-1;试管苗生根继代培养形成的试管苗生长旺盛,生根率为94.6%,生根数为7.2条,每代的繁殖系数为2.8。试管苗移栽平均成活率为89.8%,生长旺盛,在大连地区可正常越冬,翌年即可开花。 By using tissue culture technique, the tender petiole of Mahon/a bea/ei was used as explant. Researches on callus induction and differentiation, rooting of adventitious buds, rooting and transplanting of tube seed- lings, and clone establishment of Mahonla bealci were conducted in this paper. The results showed that MS + KT0.6 mg·L-1 +2,4-D 2.5 mg· L-1 was the optimmn medium for callus induction and subculture. The optimum medium for callus differentiation was 1/4MS + ZT0.4 mg · L-I + NAA 0. 1mg·L-1. After dealt with NAA (concentration of 20 mg · L-1 ) for 3 minutes, the adventitious buds could be easily induced to rooting in the medium of 1/3MS + IAA0.5 mg · L-1. The rooting rate was 94.6% with 7.2 roots on average. The surviv- al rate d transplanted tube seedlings was 89.8%. Tube seedlings could overwlnter in Dalian, growing vigorous- ly and flowering in the following year.
