还原型谷胱甘肽对急性百草枯中毒治疗作用的实验研究 被引量:7

摘要 目的 探讨还原型谷胱甘肽对急性百草枯中毒的治疗作用。方法 在大鼠用百草枯 (Paraquat ,PQ)灌胃( 2 5 0mg/kg)染毒后不同时间腹腔注射还原型谷胱甘肽 (GSH) ,分别测定染毒后 8h、2 4h、48h、72h大鼠血浆和支气管──肺泡灌洗液 (BALF)中丙二醛 (MDA)含量及超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH Px)的活力 ,并观察肺组织结构改变。结果 PQ染毒后血浆及BALF中MDA显著高于对照组 (P <0 .0 1) :血浆SOD活力下降 ,GSH Px48h后稍升高 ;BALF中SOD、GSH Px均下降 ,但前者 48h后升高 (P <0 .0 1)。给予GSH治疗后与中毒组相比 ,血浆和BALF中的MDA下降 (P <0 .0 5 ) ;SOD、GSH Px逐渐升高 ,但血浆SOD 72h似有下降。 4h、12h、2 4h三个时间点投用GSH的治疗组之间 ,SOD、MDA差异不大 ,仅 12h治疗组血浆中GSH Px稍低于 4h治疗组 (P <0 .0 5 ) ,BALF中GSH Px稍高于 4h治疗组 (P <0 .0 5 )。肺组织病理学检查可见PQ中毒后肺泡壁充血、增厚、炎症细胞浸润 ,并有局灶性出血 ;给予GSH后 ,肺泡壁充血、出血均有减轻。结论 给予还原型谷胱甘肽 。 Objective To study the therapeutic effect of reduced glutathione(GSH) against acute poisoning by paraquat. Methods SD rats were given(i.p.) GSH at different time after administration of paraquat(PQ, 250mg/kg)by oral.8h,24h,48h, 72h later, the level of MDA, and the activities of superoxide dismutase(SOD) and glutathione peroxidase(GSH Px) in plasma and bronchoalveolar lavage fluid(BALF) of the rats were detected respectively. Results The level of MDA of the PQ group significantly increased both in plasma and BALF( P <0.01) compared with control group, SOD in plasma was decreased than control group ( P <0.01), GSH Px showed some increase 48h later after poisoning. The activities of SOD and GSH Px in BALF were all decreased, but SOD showed some increase 48h later. Compare GSH treated group with PQ poisoning group, the MDA level decreased both in plasma and BALF,the SOD and GSH Px activities were increased too.The administration of GSH at different time after poisoning showed a little difference in MDA level and the activities of SOD and GSH Px.The pathological changes in lungs of PQ poisoned rats mainly showed congestion, leukocytes infiltration, and thickness of alveoli and local hemorrhage. After administration of GSH, congestion and hemorrhage of alveoli got some improvement. Conclusion Administration of GSH could improve the lipid peroxidation damage caused by acute poisoning of paraquat.
