摘要
为了进一步探讨 p CDSj32诱发小鼠抗血吸虫感染的保护免疫机制 ,分别用 10 0 μg和 30 0 μg p CDSj32质粒免疫小鼠 ,观察血清 Ig G抗体滴度的动态变化。结果显示 ,单剂量免疫后 4wk,血清 Ig G滴度可达 1∶ 32 0以上 ,以后血清抗体滴度均维持在 1∶ 6 40~ 12 80 ,最少可持续 2 4wk。末次取血清后 ,用日本血吸虫尾蚴攻击感染后 7d,测定血清抗体滴度可骤升至 1∶ 2 5 6 0以上。抗体依赖细胞介导的细胞毒试验表明 ,10 0 μg p CDSj32质粒免疫后 4wk及
In order to study the protective immune mechanisms against Schistosoma japonicum infection induced by DNA vaccine, we observed dynamics of the specific antibody IgG in sera from mice immunized with 100 μg and 300 μg plasmid pCDSj32. It was shown that IgG antibody titre was up to 1∶320 in 4 weeks after vaccination with single dose of pCDSj32. The antibody titres in the sera could be as high as 1∶640 to 1∶1 280 for more than 24 weeks afterwards. Seven days after challenge with schistosomulae, the antibody titre quickly rose higher than 1∶2 560. The trials of ADCC demonstrated that strong damage to schistosomulae could be produced by macrophages mediated with sera from mice in 4 or 8 weeks after vaccination with the DNA vaccine.
