保健灸法对高月龄大鼠抗氧化能力及脑组织中c-myc表达的影响

摘要 目的观察保健灸法对高月龄大鼠抗氧化能力及脑组织中c-myc表达的影响,以探讨保健灸法抗氧化及细胞凋亡的作用。方法 SD雌性大鼠42只,其中青年组14只,10月龄28只,随机分为对照组和保健灸组。实验两个月后采用分光光度法检测各组大鼠血清中丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性;采用免疫组化法检测脑组织中c-myc蛋白的表达水平。结果与青年组比较,12月龄对照组大鼠血清中丙二醛(MDA)含量增多,超氧化物歧化酶(SOD)活性下降,脑组织中c-myc蛋白表达水平明显升高(P<0.01)。与12月龄对照组比较,12月龄保健灸组丙二醛(MDA)含量降低,超氧化物歧化酶(SOD)活性升高,脑组织c-myc蛋白表达水平明显的下降。结论保健灸法可显著降低因增龄而造成的机体内脂质过氧化反应物的增多,具有抗氧化的功能,并且可通过调控c-myc的表达,抑制细胞的凋亡,延缓衰老。 Objective To observe the effect of health - care moxibustion on oxidation resistance and c - myc expression in months of 12 age rats and its mechansism. Methods 42 SD female rats, 14 rats in youth group and 28 rats in 10 months of age group, were randomly divide into control group and health - care moxibustion group. After two months of experiment, the content of malonaldehyde (MDA) and the activity of superoxide dismutase (SOD) in serum of rats were determined with spectrophotography; the expression level of c - myc protein in rat brain tissue was detected with immunohistochemistry. Results Compared with youth group, the content of MDA in rat serum increased in 12 months of age rats of control group, the activity of SOD was decreased, and the expression of c - myc protein in brain tissue obviously increased (P 〈 0.01 ). Compared with 12 months of age rats of control group, content of MI)A in 12 months of age rats in health - care moxibustion group was decreased ,while the activity of SOD was increased (P 〈 0.01 ), the expression level of c - myc protein in brain tissue was obviously decreased. Conclusion Health - care moxibustion can obviously inhibit the increase of product of lipid peroxidation caused by aging, and the mechanism may be related with anti -oxidation function, inhibiting apoptosis and regulating c -myc expression.
