摘要
目的比较低氧预处理和低氧后处理对大鼠心肌细胞血红素氧化酶-1水平的影响。方法将培养的心肌细胞分为四组:对照组、低氧预处理组、低氧后处理组和联合组。分别测定各时间段细胞上清液中LDH水平和HO-1水平。结果低氧预处理组和低氧后处理组LDH的水平明显低于对照组(均P<0.01),HO-1水平明显高于对照组(均P<0.05)。联合组LDH的水平及HO-1水平与低氧预处理组、低氧后处理组差异没有显著性(均P>0.05)。结论该研究结果提示低氧预处理和低氧后处理对心肌细胞的保护作用部分是通过HO-1水平增加来实现的,同时也说明二者很可能是通过同一传导通路来实现心肌细胞的保护作用。
【Objective】 To compare the effect of hypoxic preconditioning and hypoxic postconditioning on expression of heme oxygenase-1 in rat myocardial cells.【Methods】 The cultured rat cardiomyocytes were randomized into 4 groups: Control group,hypoxic preconditioning group,hypoxic postconditioning group and combination group(hypoxic preconditioning plus hypoxic postconditioning).The levels of LDH and HO-1 were determined respectively in all groups at different time.【Results】 The level of LDH was significantly lower in hypoxic preconditioning group and in hypoxic postconditioning group than in control group(both P 〈0.01),while the level of HO-1 was significantly higher than in control group(both P 〈0.05).The levels of LDH and HO-1 in combination group were not differ statistically from hypoxic pre-and postconditioning group(both P 〉0.05).【Conclusion】 The results indicate that the cardioprotection of hypoxic preconditioning and hypoxic postconditioning on myocardial cells could partly be realized by increasing heme oxygenase-1 in myocardium and probably by activating the same signal transduction pathway.
