益肾化瘀方对单侧输尿管结扎模型大鼠波形蛋白表达的影响 被引量:1

摘要 目的:动态观察单侧输尿管结扎(unilateral ureteral obstruction,UUO)模型大鼠肾组织中波形蛋白(Vimentin)的表达及益肾化瘀方对其影响,探讨Vimentin在肾间质纤维化进程中的表达变化和益肾化瘀方对其影响以及防治肾间质纤维化可能的机制。方法:将清洁级雄性大鼠150只随机分为假手术组、模型组、益肾化瘀方低剂量组、益肾化瘀方高剂量组和洛汀新组,采用单侧输尿管结扎的方法建立肾间质纤维化模型(UUO模型),分别于UUO术后第3天、第7天、第14天、第21天、第28天采用光学显微镜观察肾间质病理形态学改变,采用免疫组化(PV二步法)观察大鼠肾组织Vimentin的表达变化。结果:造模3 d后即可见肾间质水肿,随时间延长逐渐出现炎细胞浸润,部分小管消失,皮质变薄。同时,Vimentin表达增高,与时间呈相关性。用药干预后,Vimentin表达较模型组有所降低,(P<0.01或P<0.05),益肾化瘀方高剂量组与洛汀新组差异不显著。结论:益肾化瘀方组可能通过降低肾组织中Vimentin表达,减轻细胞外基质沉积,从而延缓肾间质纤维化进程。 Objective:To dynamically observe the expression of vimentin in UUO rat models and the impacts of Yishen Huayu Formula on them, in order to explore expression variations of Vimentin in the process of renal interstitial fibrosis and the impaets of Yishen Huayu Formula on it,as well as the possible mechanism of preventing and treating renal interstitial fibrosis. Methods :150 male rats with clean level were randomly divided into the sham operation group, the model group, Yishen Huayu Formula in the low-dosage group, Yishen Huayu Formula in the high-dosage group and the Lotensin group. UUO method was used to establish models of renal interstitial fibrosis, and then pathological changes in renal interstitium were observed by optics microscope in the third day, the seventh day, the fourteenth day,the twenty-first day and the twenty-eighth day after UUO operation. Expression variations of Vimentin in rats' nephridial tissue were observed by immunohistochemical method. Resuhs:Nephridial interstitial edema was seen after three days of molding. Along with time extension, infiltration of inflammatory cell gradually appeared, part tubule disappeared and cortex became thin. At the same time, Vimen- tin expression increased,which presented relativity with time. Compared with the model group, Vimentin expression in Yishen Huayu Formula of low-dosage group decreased (P 〈 O. O1 or P 〈 O. 05 ), and there was no significant difference in the Yishen Huayu Formula of high-dosage group and the Lotensin group. Conclusion : Yishen Huayu Formula can alleviate extracellular matrix deposition through decreasing Vimentin expression of innephridial tissue, so as to delay the progress of renal interstitial fibrosis.
