摘要
目的观察金匮肾气丸对异丙肾上腺素致大鼠心室重构不同阶段的影响。方法异丙肾上腺素连续3 d以20、10、5 mg/kg递减皮下注射,3 mg/kg维持7 d制备大鼠心室重构模型,造模成功后用依那普利和金匮肾气丸分别干预15周,并分别于干预5、10、15周时从心脏血流动力学、心输出量、心脏质量指数等角度评价其疗效。结果 5周时,与正常组比较,模型组左室收缩峰压(LVSP)、左室舒张末压(LVEDP)、左室内压最大收缩率(+dp/dtmax)、左室内压最大舒张率(-dp/dtmax)差异均无统计学意义(P>0.05),但降低心输出量(CO)和增加心脏指数方面差异有统计学意义(P<0.05);10、15周时,模型组LVSP、+dp/dtmax、-dp/dtmax均较正常组显著降低(P<0.05);药物干预10周后与模型组比较,金匮肾气丸能有效提高LVSP、+dp/dtmax和CO(P<0.05),并降低心脏指数(P<0.05)。药物干预15周后结果与10周相似,但是金匮肾气丸处理15周能显著降低LVEDP(P<0.05)。结论金匮肾气丸对异丙肾上腺素致大鼠心室重构早期(5周)、中期(10周)和晚期(15周)均有一定的改善作用,在中晚期时更为明显。
Objectives To observe the effect of Jingui Shenqiwan on the cardioventricular reconstruction of rats induced by Isoproterenol (ISO) in different stage. Methods The rats were injected subcutaneously by 20, 10, 5 mg/kg ISO degressively for 3 days and then 3 mg/kg ISO as maintenance dose for 7 days to build the rat model of cardioventricular reconstruction. Enalaprfl and dingui Shenqiwan were given to the rats for 5, 10, 15 weeks as interventions. The hemodynamics, cardiac output (CO) and the heart mass index were observed. Results At 5th week, the values of LVSP, LVEDP, +dp/dtma and -dp/dtma of model group had no significant differences compared with normal group (P〉0.05). The CO of model group decreased and HW/BW increased (P〈0.05). At 10th and 15th week, the values of LVSP, +dp/dtmax and -dp/dtm of model group decreased significantly compared with normal group (P〈0.05). At 10th week, compared with model group, the values of LVSP, +dp/dtmax and CO of Jingul Shenqiwan group was markedly increased (P〈0.05). Meanwhile, the value of HW/BW decreased (P〈0.05). Though the results of 15th week were similar with 10th week, LVEDP of Jinkui Shenqiwan group at 15th week was markedly deceased (P 〈0.05). Conclusion Jingui Shenqiwan has some therapeutic effects on cardioventricular reconstruction in earlier period, middle and advanced stage of rats induced by isoproterenol, and it is more obvious for middle and advanced stage cardioventricular reconstruction.
