摘要 目的 研究血管紧张素Ⅰ转换酶 (ACE)基因多态性是否与 2型糖尿病肾病 (DN)相关。方法 用聚合酶链反应 (PCR)扩增ACE基因第 1 6内含子的一个 2 87bp的插入 /缺失 (I/D)基因片段 ,1 5 %琼脂糖凝胶电泳 ,紫外线灯下观察结果。共检测了 2 2 5例江苏籍汉人 (2型糖尿病 1 75例 ,其中 1 0 0例不伴DN ,75例伴DN ;对照者 5 0例 )的ACEI/D基因型。结果 各组间ACEI/D基因型和等位基因频率分布无明显差异 (P >0 0 5 )。结论 ACE基因I/D多态性与 2型DN的发病无明显相关性。 Objective To study the relationship between angiotensin I converting enzyme (ACE) gene polymorphism and diabetic nephropathy(DN) in Type 2 diabetes.Methods A fragment of 287bp Alu sequence in inron 16 of ACE gene was used as insertion/deletion (I/D) polymorphic marker.After polymerase chain reaction(PCR) amplification of gene fragment,1 5% agarose gel electrophoresis was performed and the results were observed under ultraviolet.175 patients with Type 2 diabetes (Han,Jiangsu Province) were studied.Among them,100 who did not develop DN concomitantly formed the non DN group,while the remaining 75,having DN, made up the DN group.Control group consisted of 50 healthy subjects.Results There was no significant difference between ACE gene I/D types and frequency of allele D,I in DN and non DN group ( P> 0 05),between clinical DN and non DN group ( P> 0 05),between early DN and non DN group ( P> 0 05),between DN and control group.Conclusion There is not significant correlation between ACE gene I/D polymorphism and DN pathogenesis in patients with Type 2 diabetes.
