摘要
目的 探讨神经营养因子防止成鼠脊髓轴突损伤后引起神经元萎缩的作用。方法 采用改良的Allen’s打击法致伤大鼠脊髓 (2 5cmg) ,将实验动物分为 5组 ,A组 :单损伤组 ;B组 :损伤 +NGF组 ;C组 :损伤 +CNTF组 ;D组 :损伤 +NT 3组 ;E组 :损伤 +BDNF组。手术后应用行为学和电生理检查观察大鼠功能恢复情况 ,应用尼氏染色方法观察神经元的大小 ,采用计算机图像分析技术 ,进行定量分析。结果 神经营养因子可以防止成鼠脊髓轴突损伤后引起的神经元萎缩。图象分析发现其作用为E >D >C >B >A ,E组效果最好 ,可以较好地恢复损伤神经元的形态。大鼠神经功能的恢复也出现了相同的变化趋势。结论 神经营养因子能维持神经元的细胞形态 。
? Objective To explore the effects of neurotrophic factors on reversing the axotomy-induced neurons atrophy of injured spinal cord in adult rats. Methods The adults rats received lumbar spinal cord contusion.Experimental rats were divided into five groups:Group A:spinal cord contusion only;Group B:spinal cord contusion plus NGF;Group C:spinal cord contusion plus CNTF;Group D:spinal cord contusion plus NT-3;Group E:spinal cord contusion plus BDNF.After operation combined behavioral score (CBS),somatosensory evoked potentials (SEP),motor evoked potentials (MEP) were examined.Rats were killed after 1,3,7,14 days.Nissl stained section was used for neurons quantitative image analysis.The positive cells were quantitative analysis by a computer image analysis system. Results The cell sizes of neurons sequence were found in five groups, they are E >D> C>B>A (p<0.05).These increases in cell size of neurons were paralleled by a significant improvement in neurological function recovery. Conclusion It indicates that the neurotrophic factors can maintain the neurons morphology and improve rats hind limb function. 〔
