摘要
试验目的在于研究土壤和活性污泥的最适DNA提取方法。采用9种不同的提取方法,对所提取DNA的浓度、纯度、提取率、片段大小、16SrDNA PCR结果进行了比较,结果表明,在土壤中,液氮研磨法提取DNA最优:DNA质量浓度为800mg/L,OD260/OD280为1.7。提取率为320μg/g;在活性污泥中,液氮浸泡法提取DNA最优:DNA质量浓度为1 485mg/L,OD260/OD280为1.59,提取率为594μg/g。
The purpose of the experiment was to find the optimal DNA extraction method from soil and activated sludge.The concentration,purity,extraction rate and fragment size of DNA extracted by nine different methods and 16s rDNA PCR results were compared,and the results showed that the best extraction method of total DNA from soil was liquid nitrogen grinding:the mass concentration of DNA was 800 mg/L,OD260/OD280 was 1.70,and the extraction rate was 320μg/g.The best extraction method from activated sludge was immersion in liquid nitrogen:the mass concentration of DNA was 1 485 mg/L,OD260/OD280 was 1.59,and the extraction rate was 594 μg/g.
