PPARγ基因多态性与湖北汉族人群妊娠糖尿病的相关性研究 被引量:3

摘要 目的研究PPARγ基因单核苷酸多态性(SNPs)与湖北汉族人群妊娠糖尿病(GDM)的关系。方法测定66例GDM孕妇(患者组)及69名正常健康孕妇(对照组)PPARγ基因Pro12Ala(rs1801282)SNP多态性,分析其与GDM的关系。基因分型采用聚合酶链式反应-限制性片段长度多态性方法(PCR-RFLP)。结果患者组SNPs基因型及等位基因频率分布与对照组比较无显著性差异,患者组中rs1801282 AB+BB基因型总胆固醇(TC)、血糖和低密度脂蛋白胆固醇(LDL-C)水平显著高于AA基因型(P均<0.01);rs12636454AA基因型三酰甘油水平高于AB+BB基因型,rs11128597AA基因型血糖水平高于AB+BB基因型(P均<0.01)。结论 PPARγ基因与湖北汉族人群GDM无直接相关,但可能参与GDM的血糖水平和脂质代谢的调节。 Objective It is to study the relationship between PPARγ gene polymorphisms with gestational diabetes mellitus in Han Chinese in Hubei. Methods Sixty-six GDM women were selected as patient group, 69 normal pregnant women were se- lected as control group. The distribution of PPARγ gene Prol2Ala(rs1801282) alleles frequencies of these pregnant women were examed by polymerase chain reaction (PCR) -restriction fragment length polymorphism (RFLP) method. Results No significant difference was found in the genotypes and allelic frequencies at PPARγ 12Ala between patient group and control group. There was significant difference in total cholesterol, glucose and low density lipoprotein cholesterol between AB + BB (rs1801282) and AA (rs1801282) in GDM group (P all 〈0.01 ). The level of TG in rs12636454AA was higher than that in AB + BB in GDM group(P 〈0.05) , and the level of blood glucose in rs11128597 was higher than that in AB + BB in GDM group (P 〈0.01 ). Conclusion PPARγ gene is not directly related with GDM in Han Chinese in Hubei, but it may involves in lipid metabolism and glucose of regulation.
