六味地黄汤对非酒精性脂肪性肝病大鼠SREBP-1c和LXR-α基因表达的影响 被引量:1

Effect of Liuweidihuang decoction on sterol regulatory element binding protein-1c and liver X receptor α in the nonalcoholic fatty liver disease of rats

摘要 [目的]探讨六味地黄汤对非酒精性脂肪性肝病大鼠固醇调节元件结合蛋白(SREBP-1c)和肝X受体(LXR-α)的影响。[方法]选择雄性SD大鼠,随机分3组:正常组,六味地黄汤组(治疗组)和模型组。正常组喂养普通饲料,模型组和治疗组喂养高脂饲料建立非酒精性脂肪性肝病模型,治疗组并用六味地黄汤干预治疗;荧光定量RT-PCR检测各组大鼠SREBP-1c和LXR-α的基因表达。[结果]模型组大鼠SREBP-1cmRNA和LXR-αmRNA的相对表达均高于正常组(分别为t=2.361,P<0.05和t=3.118,P<0.01);与模型组相比,治疗组能显著降低SREBP-1cmRNA和LXR-αmRNA的表达(分别为t=3.816,P<0.01和t=3.36,P<0.01)。[结论]六味地黄汤能降低SREBP-1c和LXR-α的基因表达,改善脂质代谢。 [Objective]To explore the effect of Liuweidihuang decoction on sterol regulatory el- ement binding protein -lc(SREBP-lc)and liver X receptor a(LXR-a)in the nonalcoholic fatty liver disease(NAFLD)of rats. [Methods] Male Sprague-Dawley rats were randomly divided into model group, treatment group, normal group. To produce a fatty liver model, a high-fat diet was given for ten weeks. The normal group rats were fed normal diet. The treatment group was given Liuweidi- huang decoction while model group and normal group were given distilled water for six weeks. The expressions of SREBP-lcmRNA and LXR-αmRNA in rats liver were measured by flu- orescent quantitation reverse transcription-polymerase chain faction.[Results]The expression ofSREBP-lcmRNA and LXR-αmRNA in model group was higher than the normal group(t=2. 361,P〈0.05 and t= 3. 118, P〈0.01 respectively). The expression of SREBP-lemRNA and LXR- amRNA in treatment group was lower than that of the model group(t= 3.816, P〈0. 01;t=3.36, P〈0.01 respectively). [Conelusion] Liuweidihuang decoction could reduce the expression of SREBP-lcmRNA and LXR-αmRNA in the nonalcoholic fatty liver disease of rats.
