摘要 目的观察N-乙酰半胱氨酸(NAC)对糖尿病大鼠肾脏的保护作用,并探讨其作用机制。方法用免疫组化方法检测肾组织胶原Ⅳ、纤维连接蛋白(FN)的蛋白表达;用RT-PCR检测肾组织基质金属蛋白酶-9(MMP-9)及其组织抑制因子-1(TIMP-1)mRNA的表达。结果 NAC能减少尿蛋白,降低血肌酐水平,减少胶原Ⅳ、FN及TIMP-1mRNA的表达,增加MMP-9mRNA的表达。结论 NAC可能通过上调MMP-9在肾组织的表达,减轻细胞外基质(ECM)的聚积,起到减少尿蛋白排泄量,保护肾功能的作用。 Objective: To investigate the protective effects of NAC on the renal tissues. Methods: The following groups of rats were involved in the study: normal control rats, strep - tozotoein induced diabetic rats and diabetic rats treated with NAC. The expressions of fibronectin( FN), eollagenlV, MMP-9 and TIMP- 1 were measured by immunohistologieal and RT -PCR staining at 8 weeks of the experiment. Results: After treatment of NAC, glomerulosclerosis index(GSI) substan- tially decreased, so did the expression of collagenlV, FN and the urinary albumin excretion, ( P 〈 0.01 ), meanwhile, the expression of MMP - 9RNA was augmented, and the expressions of(TIMP - 1 ) reduced. Conclusion : NAC ean significant- ly alleviate the aggravation of renal functions in diabetic rats by means of reducing proteinuria and the accumulation of extra- cellular matrix (ECM).
