摘要
目的探讨核转录因子-κB(nuclear factor-kappa B,NF-κB)及基质金属蛋白酶(Matrix metalloproteinase,MMP)-9在两种宫颈癌siha、caski细胞中的表达及意义。方法采用免疫细胞化学法和酶联免疫吸附试验(ELISA)法检测两种宫颈癌细胞及其上清液中NF-κB及其下游调控基因MMP-9的蛋白表达。结果 NF-κB、MMP-9在宫颈癌细胞中均有表达,与siha细胞相比,两者在肠转移的caski细胞中及其上清液中的表达较高,差异有统计学意义(P<0.01)。结论在宫颈癌细胞中存在有NF-κB及其下游基因MMP-9的高表达,提示其可能在宫颈癌发生发展及转移中起重要作用。
Objective To investigate the expression and significance of NF-κB and MMP-9 in human Siha/Caski cervical carcinoma cell strains.Methods Immunocytochemistry and enzyme-linked immunosorbent assay(ELISA) were used to detect the expression of NF-κ B and MMP-9 in the two carcinoma cell strains and cellular supernatant.Results Both NF-κ B and MMP-9 were highly expressed in cervical carcinoma cells,with statistically higher expression(P0.01) in the intestine-metastasized Caski cells and their cellular supernatant compared with the non-metastasized cervical carcinoma cells Siha and their supernatant.Conclusion The high expression of NF-κB and its down-stream gene MMP-9 in cervical carcinoma cell strain may play a critical role in metastasis of the cervical carcinoma.
