海岛棉与陆地棉不同纤维发育时期苯丙烷代谢相关基因表达差异比较

摘要 【目的】比较研究在海岛棉与陆地棉纤维发育过程中苯丙烷代谢相关基因(PAL和4CL)表达模式的差异。【方法】利用荧光定量PCR,检测不同品种来源纤维在不同发育时期目的基因表达量。【结果】发现PAL1及4CL1基因的表达量随纤维发育天数的增加而增加,在陆地棉军棉1号中,基因表达量在15 DPA达到最高,而在海岛棉新海20号中PAL1及4CL1基因的表达高峰期则为20 DPA。【结论】发现PAL1基因与4CL1基因的表达模式在海岛棉与陆地棉纤维发育过程中出现差异,海岛棉中两个基因的表达高峰期均晚于陆地棉,为逐步揭示海岛棉和陆地棉纤维品质差异的分子机理奠定了一定实验基础。 [ Objective ] The purpose of this project was to investigate the differences between the expression profiles of Phenylpropanoid Pathway genes ( PAL and 4CL ) in the developing fibers between the G. hirsutum ( Junmian 1 ) and G. barbadense ( Xinhai 25 ). [ Method ] Using real - time fluorescence quantitative PCR, the target genes expression level at different fiber developing period was detected in Junmian - ! and Xinhai - 25, respectively. [ Result] The expressions of PAL1 and 4CL1 were increased along with cotton fiber development both in Junmian - 1 and XinHai - 25, but the gene expression peak appeared at 15 DPA in Junmian - 1, earlier than in Xinhai -25 (20 DPA). [ Conclusion ] The differences of gene expression profiles in the process of fiber development between the Junmian - 1 and Xinhai -25 was indicated by this experiment,and the expression peak of PAL1 and 4CL1 appeared earlier in Jumnian -1 than in Xinhai -25. This result laid an experimental foundation for the molecular mechanism research of fiber quality differences between G. barbadense and G. hirsutum.
