摘要
目的:探讨高糖高脂饮食妊娠大鼠脂联素(APN)和腺苷酸活化蛋白激酶(AMPK)mRNA的表达与胰岛素抵抗(IR)的关系。方法:将66只健康雌性SD大鼠随机分为高糖高脂饮食孕鼠组(HP组)、高糖高脂饮食未孕鼠组(HV组)、正常饮食孕鼠组(NP组)及正常饮食未孕鼠组(NV组)4组。在妊娠第20天禁食12h后测空腹血糖和胰岛素水平,计算胰岛素抵抗指数(HOMA-IR);应用RT-PCR测定脂肪组织中APN mRNA和骨骼肌组织中AMPKα2 mRNA的表达。结果:4组空腹血糖、胰岛素水平和HOMA-IR水平差异有统计学意义(P<0.001),HP组空腹血糖、胰岛素水平和HOMA-IR较其他3组明显升高(P<0.05);4组APN和AMPKα2 mRNA表达水平差异有统计学意义(P<0.001),HP组APN和AMPKα2 mRNA的表达较HV组和NP组明显降低(P<0.05),且均与HO-MA-IR呈负相关(r=-0.537,-0.516;P<0.05)。结论:APN和AMPK可能参与了妊娠期IR的发生发展。
Aim:To study the relationship between the expression of adiponectin and AMP-activated protein kinase(AMPK) and insulin resistance(IR) of pregnant rats with high sucrose and fat diet.Methods:Sixty-six healthy female SD rats were divided into four groups randomly:sucrose and fat diet pregnant rat(HP) group,sucrose and fat diet virgin rat(HV) group,normal diet pregnant rat(NP) group and normal diet virgin rat(NV) group.Twenty days after gestation,fasting plasma glucose(FPG) and fasting insulin(FINS) were measured.IR index(HOMA-IR) was calculated according to FPG and FINS.RT-PCR was performed to detect the expression of APN mRNA in adipose tissue and AMPK mRNA in the skeletal muscle tissue.Results:Levels of FPG,FINS and HOMA-IR showed significant difference in four groups(P 0.001) ;levels of FPG,FINS and HOMA-IR in HP group increased significantly than those in the other three groups(P 0.05).Levels of adiponectin mRNA and AMPK mRNA showed significant difference in four groups(P 0.001) ;levels of adiponectin mRNA and AMPK mRNA in HP group decreased significantly than those in HV and NP groups(P 0.05).The expression of adiponectin mRNA and AMPK mRNA correlated with HOMA-IR negatively in HP group(r =-0.537,-0.516;P 0.05).Conclusion:Adiponectin and AMPK might be involved in the IR in gestation.
