骨肽注射液对新生大鼠的成骨细胞增殖和碱性磷酸酶活性的影响 被引量:8

摘要 目的考察骨肽注射液对新生大鼠的成骨细胞增殖和碱性磷酸酶活性的影响。方法在体外培养的大鼠成骨细胞中分别加入75,150,300,600和1 200μg/L的骨肽注射液培养6 d,用MTT法测定成骨细胞的增殖能力;用酶联免疫法检测培养12 d的细胞内碱性磷酸酶活性。结果与对照组比,骨肽注射液150~1 200μg/L组1~6 d的细胞增长速度较快(P<0.05),并呈剂量依赖性,骨肽注射液300μg/L对成骨细胞的增殖的促进作用最为明显;骨肽注射液75~1 200μg/L组培养3,6,9和12 d的成骨细胞碱性磷酸酶活性较高(P<0.05),骨肽注射液300μg/L的碱性磷酸酶活性最强。结论骨肽注射液可提高细胞内碱性磷酸酶活性并加速成骨细胞的增殖活动。 Purpose To study the effect of bone peptide injection on neonatal rat osteoblast proliferation,and alkaline phosphatase activity.Methods Cultured rat osteoblasts were added into 75,150,300,600 and 1200 μg/L bone peptide injection respectively.Proliferation of osteoblasts 1-6 days in culture was measured by MTT.Alkaline phosphatase activity of cultured bone was collected by enzyme-linked immunosorbent assay in 6,9 and 12 days.Results Compared with control group,1-6 days,cell growth in bone peptide 150-1 200 μg/L was faster(P0.05) in a dose-dependent manner,while bone peptide 300 μg/L promoted the cell proliferation most evidently.Alkaline phosphatase activity bone peptide was higher in 75-1 200 μg/L group for 3,6,9 and 12 days of bone culture(P0.05).Alkaline phosphatase activity in 300 μg/L bone peptide was the strongest.Conclusion Bone peptide injection can increase the intracellular alkaline phosphatase activity and accelerate the proliferation of osteoblast activity.
