摘要
目的:建立血浆中维生素A的高效液相色谱快速测定方法。方法:在避光条件下,血浆加入内标后振荡混匀,经正己烷萃取后离心,取上清液氮气吹干,加入无水乙醇溶解,HPLC测定。结果:在0.034μg/ml~0.68μg/ml的浓度范围内线性关系良好,r=0.9998,方法回收率88.2%~111.8%,最低检出限0.6 ng。结论:该方法操作简便、快速、准确,能够满足日常大批量快速分析要求。
Objective:To establish a method for fast determination of vitamin A in blood plasma by high performance liquid chromatography(HPLC).Methods: Under dark condition,the plasma was shaked after adding internal standard,and centrifuged after extraction by n-hexane,then the supernatant was dried by N2 flow,dissolved wtih ethyl alcohol,and finally determined with HPLC.Results: The method had a good linearity with r=0.9998 at the concentration range of 0.034 μg/ml^0.68 μg/ml.The recoveries were between 88.2% and 111.8%.The limit of detection was 0.6 ng.Conclusion: The method was simple,fast and accurate,which can satisfy the routine heavy tasks.
