摘要
目的 研究cyclin D1基因在原发性肝细胞癌中的表达,并探讨其与P16及CDK4蛋白表达的关系。 方法 用原位杂交检测cyclin D1基因mRNA表达,免疫组化检测Cyclin D1蛋白表达。 结果 6例(14.3%)HCC可检测到cyclin D1基因过表达,5例属Ⅲ—Ⅳ级,1例属Ⅱ级。6例HCC均有CDK4蛋白表达,其中3例为P16蛋白阳性。 结论 ①cyclin D1表达增加只在小部分恶性程度高的HCC中起作用,可能与肿瘤侵袭有关。②在HCC中,cyclin D1基因过表达和(或)P16蛋白丢失均起重要作用。
AIM To study the expression of cyclin D1 gene in primary hepatocellular carcinoma, and its relationship with P16 and CDK4.METHODS In situ hybridization was used to detect the expression of cyclin D1 transcripts; and immunohistoch-emistry was employed to determine cyclin D1 protein. RESULTS Six (14.3%) of 42 HCC cases showed positive expression of cyclin D1 mRNA and its protein. Five cases were of grade Ⅲ -Ⅳ , the remaining 1 case was of grade Ⅱ. CDK4 protein was also detected in those HCCs, but only 3 cases with positive cyclin D1 showed positive P16. CONCLUSION Expression of cyclin D1 gene may play a limited role in a subset of HCC with higher tumor grade which may be correlated to the invasion of tumors. Cyclin D1 overexpresion and/or P16 protein deletion also play an important role in some HCCs.
