摘要
目的筛查胰腺组织基因mRNA的表达变化,寻找与2型糖尿病(T2DM)发生相关的基因。方法 30只SD大鼠随机分为正常对照组(A组,10只)和糖尿病模型组(B组,20只)。链脲菌素(STZ)40mg/kg联合高糖高脂饮食诱导T2DM大鼠模型,取胰腺组织常规提取总RNA,采用基因芯片检测系统进行mRNA表达谱差异分析,用实时定量PCR验证,并进行生物信息学的分析。结果基因芯片共检测出237个基因mRNA差异表达;其中,B组有17个基因发生了3倍以上的明显差异表达(上调12个,下调5个);生物信息学分析发现,差异表达基因涉及通路共9条。结论在大鼠糖尿病模型中,胰淀粉酶(Amy)1、Amy2、Amy3、聚-l-精氨酸酞酰胺(Pap2)、胰腺再生蛋白3α(Reg3a)等基因的表达均发生明显变化,提示其可能与糖尿病的发生密切相关。
Objective To screen the changes of gene mRNA expression in the pancreas and search for type 2 diabetes mellitus(T2DM)-related gene. Methods Thirty SD rats were randomly divided into two groups of A(normal controls, 10 rats) and B(diabetes mellitus model, 20 rats). The T2DM rat model was established by streptozocin(STZ) 40 mg/kg plus high-sugar and high-fat diet. The rats were sacrificed and total RNA was extracted from the pancreas. The differential analysis of mRNA expression profile was performed by gene Chip and validated by real-time PCR and the related pathways were analyzed by bioinformatics. Results The differential expression of 237 gene mRNAs was detected by gene chip. The differential expression more than triple was found in 17 gene mRNAs in group B, of which 12 gene mRNAss were up-regulated and the other 5 down-regulated. Bioinformatics showed that 9 pathways were related to the differential expression of 237 gene mRNAs. Conclusion The expressions of Amyl, Amy2, Amy3, Pap2, Reg3a and 6ther genes are significantly changed in the pancreas of T2DM rats, indicating that those genes may play an important role in the development of diabetes mellitus.
