FHIT、FN和PTEN在肝癌组织中的表达及临床意义 被引量:14

Expression of FHIT, FN and PTEN in hepatocellular carcinomas

摘要 目的探讨脆性组氨酸三联体基因蛋白(fragile histidine triad,FHIT)、纤黏连蛋白(fibronectin,FN)、张力蛋白同源基因蛋白(phosphatase and tensin homology deletedo n chromosome ten,PTEN)在肝癌组织中的表达及与患者预后的关系。方法应用免疫组化方法检测138例肝癌组织标本的FHIT,FN,PTEN的表达情况,分析表达与患者临床病理因素的关系。差异检验采用Log—rank检验。结果FHIT,FN,PTEN在肝癌组织中的表达与癌旁组织比较差异有统计学意义(分别χ2=5.968,7.380,4.962,均P〈0.05),与肿瘤大小,分化程度,肿瘤数目,淋巴结转移,癌栓形成等比较差异有统计学意义(P〈0.05),FHIT,FN表达阳性患者组较阴性患者组生存时间短(分别χ2=4.443,9.867;均P〈0.05),而PTEN表达阳性患者较阴性患者生存时间长(分别x。=4.199,P〈0.05)。结论FHIT,FN,PTEN在肝癌组织中存在异常表达,FHIT,FN表达提示患者预后不良,PTEN表达提示患者预后较好。 Objective To investigate the expression of fragile histidine triad ( FHIT), fibronectin (FN) and phosphatase and tensin homology deleted on ehromosome ten (trFEN)in human hepatocellular carcinomas (HCC) and their relationship with pathological characteristics and prognosis of HCC. Methods Immunohistochemistry was used to detect expression of FHIT, FN and PTEN in cancerous tissues from 138 HCC patients. The correlation between their expression and clinicpathological features and prognosis were analyzed. Results FHIT, FN, PTEN protein expressed differently between HCC and adjacent mueosa (respectively χ2 = 5. 968, 7. 380, 4. 962, all P 〈 0. 05), and the expression level was different with tumor size, tumor number, grades and tumor lymph node metastasis ( P 〈 0. 05 ). In FHIT and FN positive group, the recurrence free survival rates were lower than those in negative groups( respectively χ2 = 4. 443, 9. 867, all P 〈 0. 05 ), while in PTEN positive group patients' recurrence free survival rates were higher than those with PTEN negative expression (χ2 = 4. 199, P 〈 0.05 ). Conclusions FHIT, FN, PTEN were abnormally expressed in HCC. Positive expression of FHIT and FN predicts poor prognosis, while positive PTEN indicates fair prognosis.
