摘要
用二阶导数紫外光谱法测定血清茶碱浓度,本法简便、快速、重现性好,适合作临床常规的茶碱血药浓度监测用。血清用氯仿一无水乙醇(3:1)提取。有机相直接作二阶导数光谱测定,入294nm 处d^2A/△入~2对零点的振幅D 与浓度非常相关,r=0.9996,直线方程为D=1.38c+0.74。方法的平均回收率为100.37±4.78(均值±标准差),差异系数为4.76%。以临床血样与用荧光偏振免疫测定法平行测定进行比较,结果非常一致,平均偏差0.43(μg/ml)。
The second derivative UV-spectrophotometric method to determine theophyllinein serum is simple,rapid、reproducible and suitable for theophylline monitoringin routine.The serum is extracted with chlorform-absolute alcohol(3:1)andthe organic phase is determined.At λ=294nm there is a good correlation betweenthe amplitude of d^2A/λ~2 from zero line to the curve top(D)and concentration oftheophylline(C),γ=0.9996,D=1.38C+0.74.The average recovery and coefficientof variation are 100.37±4.78 and 4.76% respectively.As compared with Fluoresce-nse Polarization Immunoassay for same serum sample from patients used aminoph-ylline,the results of determination between two methods are in good agreement(average derivation=0.43μg/ml).
