摘要
目的研究单纯疱疹病毒1型(Herpes simplex virus type 1,HSV-1)基因转移载体在BALB/c小鼠体内的生物分布。方法建立实时定量PCR反应方法,并对方法进行验证。BALB/c小鼠经肌肉注射1×107pfu HSV-1,每隔2 d给药1次,共3次。分别于末次给药后24 h和63 d摘取组织脏器,采用建立的实时荧光定量PCR法检测不同组织脏器中HSV-1基因拷贝数。结果建立的荧光定量PCR法线性范围宽,特异性强,重复性好。HSV-1基因转移载体广泛分布于各个组织器官,主要分布在背根神经节、注射部位、肺和脾脏,分布量随注射时间的延长逐渐下降。结论 HSV-1经肌肉注射后,能高效地将目的基因转运到小鼠体内的各个部位;HSV-1能为基因转移提供安全,有效的载体平台。
Objective To investigate the biodistribution of herpes simplex virus type 1(HSV-1) gene transfer vector in BALB / c mice.Methods A real-time fluorescent quantitative PCR method was developed and verified.BALB / c mice were injected i.m.with HSV-1 at a dosage of 1 × 107 pfu,once 2 days for 3 times,of which the tissues and organs were isolated and determined for HSV-1 gene copy number by the developed real-time fluorescent quantitative PCR method.Results The developed real-time fluorescent quantitative PCR method showed wide linear range,high specificity and high reproducibility.HSV-1 gene transfer vector was distributed widely in various tissues and organs,mainly in dorsal root ganglion(DRG),injection site,lung and spleen,of which the copy number decreased with the increasing time after injection.Conclusion HSV-1 delivered by intramuscular injection transferred the target gene to various tissues or organs of mice effectively,which provided a safe and effective vector platform for gene transfer.
