姜黄素对慢性脑缺血老龄鼠认知功能障碍和血清HDL水平的影响

Effect of curcumin on cognitive impairment and serum HDL level of aged rats with chronic cerebral ischemia

摘要 目的观察姜黄素对慢性脑缺血老龄鼠认知功能障碍和血清高密度脂蛋白(High density lipoprotein,HDL)水平的影响。方法采用双侧颈总动脉永久性结扎(Permanent occlusion of bilateral common carotid arteries,2VO)建立老龄鼠慢性脑缺血模型,并将模型鼠随机分为单纯缺血组以及姜黄素高剂量组(100 mg/kg)和低剂量组(50 mg/kg),另设假手术组和正常对照组,单纯缺血组、假手术组和正常对照组注射等量DMSO。各组均于术后24 h开始经腹腔注射,每日1次,连续30 d。采用Morris水迷宫行为学实验测试老龄鼠的空间学习及记忆功能,全自动生化分析仪检测血清HDL水平。结果给药30 d后,经连续5 d的训练,姜黄素高、低剂量组的逃避潜伏期均明显短于单纯缺血组(P<0.01),在原平台所在象限停留时间占总游泳时间的比例明显高于单纯缺血组(P<0.01或P<0.05),血清中HDL水平明显高于单纯缺血组(P<0.01)。结论姜黄素可以改善慢性脑缺血老龄鼠的认知功能障碍,并提高血清HDL水平。本实验为早期诊断血管性痴呆提供了新的生物学指标,也为姜黄素治疗血管性痴呆提供了新的药物靶点。 Objective To observe the effect of curcumin on cognitive impairment and serum high density lipoprotein(HDL) level of aged rats with chronic cerebral ischemia.Methods Rat model of chronic cerebral ischemia was established by permanent occlusion of bilateral common carotid arteries(2VO).The model rats were randomly divided into five groups,and those in high and low dose curcumin groups were injected i.p.with curcumin at dosages of 100 and 50 mg / kg respectively,while those in ischemia,sham operation and normal control groups with DMSO each at an equal volume,24 h after operation,once a day for 30 d,then assessed for spatial learning capacity and cognitive function in Morris water maze,and determined for serum HDL level by full-automatic biochemical analyzer.Results As compared with those in ischemia group,the escape latency periods of rats after treatment with 100 and 50 mg / kg curcumin for 30 d and being trained for 5 d were shortened significantly(P 〈 0.01),while the percentage of time in target quadrant were significantly high(P 〈 0.01 or P 〈 0.05),and the serum HDL levels were significantly high(P 〈 0.01).Conclusion Curcumin improved the cognitive impairment and increased the serum HDL level of aged rats with chronic cerebral ischemia.The study provided a new biological index for early diagnosis of vascular dementia and a new target for treatment of the disease with curcumin.
