摘要
目的研究鲨鱼软骨粉提取物对大鼠肺微血管内皮细胞的影响。方法用流式细胞仪(PI染色)测定体外培养的肺微血管内皮细胞中细胞周期和细胞凋亡的变化。结果体外培养的微血管内皮细胞与鲨鱼软骨粉全成份(6mg/ml)、成份A(0.01mg/ml)、成份B(0.01mg/ml)一起培养24h后经流式细胞仪检测发现 ,成份B使细胞周期中G2-M期细胞明显减少(P<0.05) ,成份A使凋亡细胞明显增加(P<0.01)。结论成份B可能是鲨鱼软骨粉中的一种水溶性抑制成份 ,它对微血管内皮细胞的增殖起抑制作用 ,成份A对细胞凋亡的作用 。
Objective To study the effects of shark cartilage and its extracts on pulmonary microvascular endothelial cells in rats. Methods Using cytoflowmeter method (PI stain) to detect the change of cell cycle and cell apoptosis in cultured microvascular endothelial cells. Results 24 hours after culture the element B induced decrease of cell percentage of G2-M phase in cell cycle (P<0.05) and element A induced increase of apoptosis (P<0.01) significantly using cytoflowmeter. Conclusion The element B probably is a soluble inhibitor in shark cartilage and it may inhibit the proliferation of microvascular endothelial cells. The effect of element A on the cell apoptosis is uncertain at present. Further study is necessary.
