摘要
目的:采用高效液相色谱(HPLC)法测定裸花紫珠中毛蕊花糖苷的含量。方法:采用ZORBAX SB C18(4.6 mm×250 mm,5μm)色谱柱,流动相为0.1%磷酸溶液:乙腈(77∶23),流速1.0 mL.min-1,柱温30℃,检测波长为334 nm,进样量取10μL。理论板数为6 193,分离度为4.1。结果:毛蕊花糖苷浓度在10.18～402.39μg(r=0.999 9,n=7)范围内线性关系良好,平均回收率(n=6)为96.13%,RSD=1.38%。结论:本研究方法专属性强、重复性好、结果准确,为裸花紫珠中毛蕊花糖苷含量测定提供了科学、专属的方法。
OBJECTIVE： To determine the content of verbascoside in callicarpa nudifloraby HPLC.METHODS： The chromatographic column ZORBAX SB C18（4.6 mm×250 mm,5 μm） was used with column temperature maintained at 30 ℃.The mobile phase consisted of 0.1% phosphoric acid solution-acetonitrile（77∶23） with flow rate of 1.0 mL·min-1,detection wavelength of 334 nm and sample size of 10 μL.The number of the theoretical plates were 6193 and the resolving power was 4.1.RESULTS： The linear range of verbascoside was 10.18-402.39 μg（r=0.999 9,n=7） with an average recovery rate of 96.13%（n=6,RSD=1.38%）.CONCLUSION： The method is specific,reproducible and accurate in result,and it is a scientific and specific method for determining verbascoside content in callicarpa nudiflora.
