内皮细胞特异性SIRT1过表达抑制高血糖诱导的血管细胞衰老 被引量:2

摘要 SIRT1作为Ⅲ类组蛋白去乙酰化酶能够降低高糖诱导的内皮细胞衰老,并抑制高血糖引起的血管功能失常.然而,在体内SIRT1能否抑制高血糖诱导的血管细胞衰老目前尚不清楚.应用内皮细胞特异性SIRT1转基因小鼠和野生型小鼠构建40周STZ诱导的糖尿病模型,42.9%野生型小鼠和38.5%SIRT1转基因小鼠构建成功.40周高血糖刺激成功诱导了小鼠主动脉衰老的发生,表现为衰老标志物p53,p21及PAI-1表达的显著上调.但与野生型糖尿病小鼠相比,内皮细胞特异性SIRT1过表达使得这些衰老标志物表达明显下调.另外,高血糖使得野生型小鼠主动脉中MnSOD水平显著下降,而SIRT1内皮细胞特异性过表达使其水平得以保持.此外,p66Shc作为氧化应激接头分子在糖尿病小鼠主动脉中被明显诱导上调而在SIRT1转基因糖尿病小鼠主动脉中这种上调被显著抑制.结果表明,SIRT1可以抑制高血糖诱导的血管细胞衰老的发生,至少部分是通过下调氧化应激实现的.
