摘要 目的比较肝损伤患儿与健康对照儿童之间血清代谢组的差别,从中进一步寻找可能的生物标记物。方法 30例肝损伤患儿和30例正常对照儿童分别留取血清样本,经前处理后采用高效液相色谱-串联质谱(HPLC-MS/MS)技术采集数据,并用PCA法联合t检验分析处理数据。结果肝损伤患儿与正常对照儿童在PCA模型中的数据分布差异具有统计学意义。研究共发现23种潜在生物标记物。结论应用代谢组学方法可以反映儿童肝损伤的存在,并可以通过此方法寻找儿童肝损伤新的生物标记物。 Objective To compare the serum metabolic profiles between patients and control, and thus identify the potiential biomarkers. Methods Thirty patients with infectious liver damage and 30 controls were enrolled. High performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) was used for analysis of serum sample. PCA combined with t-test was used to compare the distribution of patient group with control group. Results The distribu- tion of patient was well separated from that of control group in PCA model, which indicate there is difference significant in metabolic profile between two groups, and 23 biomarkers were identified. Conclusions Metabolomics can indicate the children's infectious liver damage and be successfully used in discovering biomarkers.
