白细胞介素-17在系统性红斑狼疮发病机制中的作用初探 被引量:1

摘要 目的探讨白细胞介素(interleukin,IL)-17在系统性红斑狼疮(systemic lupus erythematosus,SLE)中的表达情况及其与临床指标的相关性。方法采用酶联免疫吸附法检测SLE和健康对照组之间血浆IL-17表达的差异,采用实时荧光定量-聚合酶链反应检测IL-17和维甲酸相关孤儿受体(retinoic acid-related orphan receptor-γt,RORγt)的mRNA基因表达,并且分析它们之间的相关性。结果与比较健康对照组比较,SLE患者组血浆中IL-17浓度明显升高,血浆IL-17和血沉、C反应蛋白和SLE疾病活动性指数(systemic lupus erythematosus disease activity index,SLEDAI)之间具有正相关性,但与免疫指标IgG、补体之间没有明显相关性。SLE患者组IL-17 mRNA和RORγt mRNA的表达也较对照组升高,并且与血浆中IL-17的浓度呈正相关。结论 SLE中IL-17的表达水平明显异常,并且与调控基因RORγt mRNA的表达具有一致性。 In order to study the expression of interleukin(IL)-17 in systemic lupus erythematosus (SLE) and investigate the relationship between IL-17 and clinical index, we detected IL-17 by enzyme-linked immunosorbent assay (ELISA), IL-17 mRNA and retinoic acid-related orphan receptor-γt (RORγt) mRNA by real time fluorescent quantitation polymerase chain reaction (RT-PCR) in 20 cases of SLE and 15 cases of normal control, and then analyzed the relationship between them by statistical analysis. The results showed the level of IL-17, IL-17 mRNA and RORγt mRNA were all increased significantly in SLE patients compared with normal controls. The levels of IL-17 expression in SLE patients were positively associated with ESR, CRP, systemic lupus erythematosus disease activity index (SLEDAI) and RORγt mRNA, respectively. But IL-17 was not correlated with IgG, C3 or C4. All this result suggested the expression of IL-17 is abnormal in SLE, and there is concordance with its transcription gene RORγt mRNA.
