胰岛素受体底物1与肝硬化胰岛素抵抗的研究进展 被引量:1

摘要 肝硬化与胰岛素抵抗密切相关,临床表现为高糖血症、高胰岛素血症、高脂血症以及各种细胞因子的异常增高,其病理机制目前尚不明确。近来研究发现,肝硬化胰岛素抵抗与胰岛素受体底物有关,而胰岛素受体底物是胰岛素信号传导通路中的关键信号分子,它可通过表达上调或下调、翻译后修饰,以及亚细胞定位来改变胰岛素信号的传导,其中翻译后修饰中的磷酸化/去磷酸化显得尤为重要且较为复杂。再者,不同病因的肝硬化胰岛素受体底物的作用途径并不相同。 Liver cirrhosis(LC) and insulin resistance( IR hyperglycemia, hyperinsulinism, hyperlipidemia and high ) are closely correlated, clinically presenting cytokines levels, however, the underlying mechanism is not completely clear. Recent reports show that insulin receptor substrate-1 ( IRS-1 ) is associated with IR in LC. IRS-1 plays a pivotal role on insulin signal transduction; it changes insulin signaling by up- or down-regulating of protein presentation, post-translational modification and subcellular localization of proteins, particularly in phosphorylation/dephosphorylation of post-traslational modification. Furthermore, LC with different etiology may have different mechanism of IRS-1 effect on IR.
