摘要
目的:探索抗逆转录病毒疗法(ART)治疗在HIV-1疾病进程中对调节性T细胞(Treg细胞)的影响,并探讨Treg细胞频率在HIV-1疾病进程中的作用。方法:抽取114例(男96例、女18例)HIV-1阳性患者及17例健康对照者外周血,应用流式细胞术检测Treg细胞,并分析其表达水平(频率和绝对数)在HIV-1疾病进程中的变化趋势及其与CD4+细胞绝对数之间的相关性。结果:随着HIV-1感染者病情进展,患者外周血中Treg细胞绝对数趋向下降并且与CD4+T细胞绝对数呈正相关,而Treg细胞频率趋向升高并且与CD4+T细胞绝对数呈负相关。Treg细胞频率及绝对数在ART治疗无症状HIV-1阳性感染者中显著降低,而在AIDS患者中却显著升高。结论:Treg细胞参与艾滋病免疫发病过程,并且在HIV-1感染的不同阶段,ART治疗对Treg细胞水平具有一定的影响,提示通过控制Treg细胞的水平可能有助于HIV-1感染疾病的临床控制。
Objective: To understand the changes of CD4 FoxP3 regulatory T cells during disease progression of HIV-1 infection with anti-retroviral therapy (ART). Methods: 114 patients HIV-1 positively infected and 17 healthy controls were randomly enrolled. The percentages and absolute counts of Treg cells were detected by using flow cytometry and the correlation of Treg cells with the absolute CD4 T cells was analyzed in the process of disease progression of HIV-1 infection with ART. Results: With HIV-1 disease progression, the absolute counts of Treg cells in HIV-1 positive peripheral blood tended to decrease and positively correlated with the absolute counts of CD4 T cells, while the frequency of Treg cells increased and negatively correlated with the absolute counts of CD4 T cells. Both the frequency and absolute number of Treg ceils in HIV-1 positive infected asymptomatic patients with ART treatment were significantly reduced, on the contrary they were significantly higher in patients with AIDS. Conclusion: Treg cells are involved in the immune pathogenesis of AIDS. ART treatment and gender differences affect the levels of Treg cells in different stages of HIV infection. This suggests that the regulation of the levels of Treg cells may contribute to the clinical control of disease.
