苯并(a)芘对绒癌细胞系JEG-3细胞周期阻滞和分泌功能的影响

Effects of Benzo(a)pyrene on Cell cycle Arrest and Secretion of JEG-3 Choriocarnoma Cells in Vitro

摘要 目的探讨苯并(a)芘[B(a)P]对体外培养绒癌细胞系JEG-3的影响。方法不同浓度B(a)P(0.500、1.000、2.000、4.000、8.000μmol/L),染毒24h,分别应用流式细胞术和免疫化学发光法研究B(a)P对体外培养的JEG-3细胞周期和细胞因子人绒毛膜促性腺激素(hCG)分泌情况的影响。结果 JEG-3在4μmol/L和8μmol/L浓度组G2期细胞数量,与对照组相比分别上升了6.5%和21.0%(P<0.01)。随着B(a)P染毒浓度的升高,hCG分泌呈先上升后下降的趋势,2.000μmol/LB(a)P染毒组分泌达到峰值,随后降低,与对照组相比有统计学意义(P<0.05),呈现剂量-效应关系。结论 B(a)P可以抑制细胞DNA合成,引起G2期细胞阻滞,使细胞有丝分裂延迟,并且通过影响hCG的分泌,从而调节细胞的功能。 Objective To study the effect of benzo(a) pyrene[ B( a)P] on cells cycle arrest and secre- tion of JEG-3 choriocamoma ceils in vitro. Methods Ceils at exponential growth phase in vitro were treated with B (a) P at different concentrations ( 0.500,1. 000,2. 000,4. 000,8. 000 μmol/L) for 24h, respectively. The pereentage of ceils in the each phase of cell cycle was analyzed by the flow cytometry, and the supematants were collected and analyzed by immunochemiluminometric assay to quantify the release of hCG from the cells. Results In the JEG-3 cells,the decreased cell proliferation rate in all treatment groups was observed (P 〈0.01 ). The accumulation of cells in G2 phase of the cell cycle was found in 4. 0001μmol/L and 8. 0001μmol/L treatment groups. The hCG secretion was first increased and then showed a downward trend with the increase of B (a) P concentrations,which reached the highest point at the concentration of 2.000μmol/L. Compared with the control group, there was statistically significant ( P 〈 0.05 ). Conclusion B (a) P can inhibit DNA synthesis, induced the blockage G2, phase delay the mitosis of ceils, and influence the secretion of hCG, thereby regulate cell function.
