摘要
目的研究TNF-α对人牙周膜细胞(hPDLCs)表达MMP-9的影响。方法分别以1、2、5 ng/mL浓度的TNF-α刺激hPDLCs 24 h,以及2 ng/mL浓度的TNF-α刺激hPDLCs 6 h、12 h、24 h、48 h,应用ELISA法测定细胞上清液中MMP-9的表达水平。结果随着TNF-α浓度增大和作用时间延长,hPDLCs表达MMP-9均增加。结论 TNF-α能够以时间和剂量依赖的方式增强hPDLCs表达MMP-9。
Objective To examine the effects of tumor necrosis factor (TNF-α) on the regulation of MMP-9 expression in human periodontal hgment cells (hPDLCs). Methods hPDLCs were treated with different concentrations (1,2,5 ng/mL) of recombinant human TNF-α (Boster,Wuhan) for 24 h,and were stimulated by 2 ng/mL TNF-α for 6 h, 12 h,24 h and 48 h,respectively. MMP-9 protein levels in cultured supematants were determined by enzyme-linked immunosorbent assay. Results Compared with the control group,a significant increase in MMP-9 expression was detected after TNF-α stimulation with the increase of the action time and concentration. Conclusion TNF- α is a potent inducer of MMP-9 secretion in a dose- and time-dependent manner.
