农杆菌介导BADH耐盐基因转化马铃薯的研究 被引量:6

摘要 以马铃薯品种费乌瑞它微型薯为外植体,采用农杆菌介导法将BADH耐盐基因导入马铃薯中,优化了影响遗传转化的因素,包括激素浓度、脱菌抗生素浓度、共培养时间、筛选剂浓度等。共获得30株PPT抗性植株,PCR检测10株为阳性植株,Southern检测4株为阳性植株。生理检测表明植株在0.7%NaCl浓度下耐盐性相对较高。证明外源基因已经整合到植物的基因组上。 BADH gene was transferred into the potato cultivars "Favorite" by Agrobacterium-mediated transformation,and influence factors of genetic transformation were optimized.Microtuber discs were used as explants.30 resistant plants of "Favorite" were obtained.Resistant plants were blot analysis by PCR and Southern blot,of which 10 plants and 4 plants had a positive amplification.The main purpose of the studies was the identification of salt-tolerance in the potted plant with BADH gene had relatively higher salt tolerance under the concentration 0.7% NaCl stresses.The results showed that single copies of the BADH gene were inserted into the genome of the transgenic plants.
