L-肉碱的合成研究

摘要 以麦芽糖为原料,一步法合成了中间体——(S)-3-羟基-γ-丁内酯,产率为31%;再经开环、手性转化制备了L-肉碱,产率为54%。同时考察了底物、浓度、温度及时间等影响反应的因素。该合成路线避免使用剧毒物质氰化钠、外消旋体的拆分等难题,缩短了反应步骤。 (S)-3-hydroxy-γ-butyrolactone was synthesized from maltose in one step, and the yield was 31%. L-carnitine was then obtained through the ring-opening reaction, a chiral inversion, and the overall yield was 54%. Influential factors upon the reaction such as reaction substrate, concentration, temperature, and time were investi- gated. Difficult problems such as the usage of an extremely poisonous NaCN and the resolution of a racemic modification were avoided, and operational processes were shortened through this synthetic route.
