摘要
目的研究盐酸益母草碱对缺糖/缺氧损伤大鼠肾上腺嗜铬瘤细胞株(PC12)的保护作用。方法采用连二亚硫酸钠(Na2S2O4)+缺糖建立PC12细胞缺糖/缺氧损伤模型(oxygen and glucose deprivation,OGD),MTT法检测各组细胞存活率,流式细胞仪检测各组细胞内活性氧含量。结果与模型组相比,25μmol·L-1盐酸益母草碱显著提高细胞存活率,降低细胞内的活性氧含量。结论盐酸益母草碱对PC12细胞缺糖/缺氧损伤模型有明显的保护作用。
Objective To study the protective effect of the hydrochloride leonurine on deprivation of oxygen/glucose injury in rat adrenal chromium tumor cell line (PCl2). Methods We used the sodium dithionite (Na2S2O4 ) + sugar-free in PC12 ceils to bulid glucose deprivation/hypoxia injury model (the oxygen and glucose, deprivation, OGD). Cell viability was measured by MTF assay, flow cytometry detection was used to determine the oxygen content of active cells in each group. Results Compared with model group, the 25 μmol ·L^-1hydrochloride leonurtne significantly improved cells viability,reducing intracellular reactive oxygen species. Conclusion The hydro- chloric leonurine has a significant protective effect on PCl2 cell hypoglyeemia/hypoxia injury model .
