摘要
目的:建立测定茯苓药材中茯苓酸含量的反相高效液相色谱法。方法:采用HPLC法。色谱柱为Diamonsil C18柱(250mm×4.6mm,5μm),流动相为甲醇-水溶液梯度洗脱,流速为1.0mL/min,柱温为30℃,检测波长为250nm。结果:茯苓酸进样浓度在50~3 000μg/mL之间有良好的线性关系。平均加样回收率为98.53%,RSD(n=6)为2.24%。结论:HPLC法可作为茯苓质量控制的一个有效方法。
Objective:To develop HPLC for the determination of pachymic acid in Poria(the dry sclerotium of Poria cocos(schw.)Wolf.).Methods:The separation was performed on a Diamonsil C18 column(250mm×4.6mm,5μm) with methanol-water as themobile phase at a flow rate of 1.0mL/min.The detective wavelength was set at 250nm and the column temperature was 30℃.Results:There were good linear relationships between the concentrations and the peak areas of pachymic acid in the range of 50~3000μg/mL(R2=0.999 5).The recoveries were found to be 98.53% with RSD of 2.24%(n=6).Conclusion: This meothod is suitable for the quality control of Poria.
