老年学习记忆损害大鼠海马和新皮质小白蛋白神经元的丢失 被引量:7

摘要 目的 探讨老年学习记忆损害与海马和新皮质 GABA能神经元亚型——小白蛋白 (PV)神经元的关系。方法 通过Morris水迷宫行为检测将相同年龄老年大鼠分为老年学习记忆正常和损害两组 ,采用免疫组化方法定量计数各脑区 PV神经元数密度。结果 老年记忆损害鼠 CA3区和齿状回 PV神经元数密度较老年正常鼠明显减少 ;PV神经元数密度与大鼠在水迷宫中的行为密切相关。结论  CA3区和齿状回 PV神经元丢失可能是老年记忆损害的原因之一 ,提示 GABA能系统可能参与老年学习记忆损害。 To study the protective effects of intravenous basic fibroblast growth factor(bFGF) with different concentratin against acute injury following 3 hours of focal cerebral ischemia and reperfusion.Methods The model of focal cerebral ischemia/reperfusion was established by intraluminally sutring the right middle cerebral artery of mature Sprague Dawley rats.BFGF(5,50,250 μg/kg/h) was infused by femoral vein for 3h,beginning during reperfusion after 3 hours of focal inchemia.After 24h,the rats were sacrificed and the volume of cerebral infarcts was determined by TTC staining and image analysis.Results We found that the volume of cerebral infarcts(contral without NS,25 66±1 89%,with NS,23 23±2 64%;bFGF 5 μg/kg/h,14 69±2 43%;bFGF 50 μg/kg/h,10 98±3 10%,bFGF 250 μg/kg/h,13 57±3 43%,mean±SEM) was less in rats treated with bFGF,and the infart volume was less in rats treated with bFGF 50 μg/kg/h compared with bFGF 5 μg/kg/h.Conclusion There data indicated that intravenous adiminstration of bFGF could ameliorate significantly acute injury resulting from focal cerebral ischemia and reperfusion,and there was dose dependent effect.bFGF may provid its beneficial effect by limiting progression of injury in ischemic broder regions.
