补锌对衰老大鼠睾丸细胞凋亡和一氧化氮合酶的影响 被引量:5

摘要 目的 探讨补锌对衰老过程睾丸细胞凋亡的影响及其可能机制。方法 选用 2 4月龄 Wistar雄性大鼠 1 6只 ,均分为实验组和对照组 ,实验 (补锌 )组和对照组分别喂养补锌和常锌饲料。喂养 4 w后 ,采用原子吸收分光光度法测定血清锌含量 ;采用放射免疫分析法测定血清睾丸酮含量 ;采用 NADPH- d组织化学法显示睾丸细胞一氧化氮合酶 (NOS)活性 ;采用原位缺口平移技术 (INST法 )显示睾丸凋亡细胞数目。结果 与对照组相比 ,老年大鼠补锌 4w后 ,血清锌含量明显升高 (P<0 .0 5) ;血清睾丸酮水平明显升高 (P<0 .0 5) ;睾丸间质细胞 NOS阳性细胞数目明显减少 (P<0 .0 1 ) ,NOS阳性细胞反应强度明显减低 ;睾丸生精细胞凋亡数目明显减少 (P<0 .0 5)。结论 锌是睾丸发育必需的营养素 ,补锌可使老年大鼠睾丸细胞凋亡数目减少 ,这种变化可能与补锌对衰老大鼠睾丸细胞 NOS活性和睾丸酮水平的调节有关。 To study the effective mechanism of zinc supplement on testis apoptosis.Methods 16 wistar male rats(24 months) were evenly divided into zinc supplement and control groups after 4 weeks,and the serum zinc levels and testosterone contents were measured.The apoptosis of spermatogenic cells were studied in situ with nick translation(ISNT) technique.Results As compared with the control group,the serum zinc and testostrone contents were obviously increased(P<0 05),NOS positive cell in testis was significantly decreased(P<0 01) and apoptosis number was obviously decreased in zinc supplement group.Conclusions Adequate amouunt of zinc supplement can decreased testis apoptosis of aging rats,but the serum testostrone increased and NOS positive cell with decreased activity might be one of the importent mechanism of zinc supplement which can inhibit testis apoptosis of the aging rats.
