摘要 目的 探讨血管内皮生长因子 (VEGF)在膀胱肿瘤发生发展中的作用。 方法 采用逆转录聚合酶链反应 (RT PCR)方法检测三种人膀胱癌细胞系 (BIU 87,T2 4,EJ)和正常膀胱组织中VEGFmRNA的表达 ,同时用免疫细胞化学方法检测三种人膀胱癌细胞系VEGF蛋白的表达。 结果 三种膀胱癌细胞系均有VEGFmNRA的表达 ,正常膀胱组织无表达 ;免疫细胞化学检测显示 ,VEGF蛋白表达于三种细胞系 ,而以EJ和T2 4表达较强 ,BIU 87表达较弱。 结论 VEGF在膀胱癌细胞系中的表达提示其可能在膀胱肿瘤的发生和发展中起重要作用 ,它可以刺激膀胱肿瘤的血管生成 ,促进肿瘤的生长。 Objective To explore the role of VEGF in the occurrence and progression of bladder cancer. Methods Expression of VEGF mRNA was examined with reverse transcriptase polymerase chain reaction (RT PCR) in 3 kinds of human bladder tissues.The expression of VEGF protein was assayed with immunocytochemical technique. Results Expression of VEGF mRNA was detected in the bladder cancer cell lines but not in the normal bladder tissues.All the 3 bladder cancer cell lines showed positive staining of VEGF by means of immunocytochemical technique.Expression of VEGF was strong in EJ and T24 cell lines but weak in BIU 87 cell line. Conclusions Expression of VEGF in bladder cancer cell lines may be related to the occurrence and progression of bladder cancer.VEGF could induce tumor angiogenesis and would accelerate the tumor growth.
