摘要
目的 应用免疫组化法 (ABC法 )检测 ICA,比较研究 A型、B型、AB型与 O型 4种不同血型成人胰腺抗原底物对 ICA检测的特异性和敏感性的影响。方法 取 2 4例 1型糖尿病病人的 ICA阳性不同 JDF单位的血清标本和 2 3例正常人 ICA阴性血清标本 ,以 A型、B型、AB型与 O型 4种不同血型成人胰腺的冰冻切片为抗原底物 ,应用 ABC免疫组化法检测 ICA。结果 以 O型血成人胰腺为对照 ,应用 3例 A型和 B型血胰腺和 2例 AB型血胰腺为抗原底物进行ICA检测 ,2 4例 ICA阳性血清和 2 3例 ICA阴性血清检测结果的特异性和敏感性基本一致 ,未见假阳性或假阴性结果。结论 本文研究结果证明 A型、B型或 AB型血成人胰腺与 O型血成人胰腺一样 ,可用于 ICA免疫组化 (ABC法 )检测 ,不同血型人胰腺作为抗原底物不影响 ICA免疫组化法检测的特异性和敏感性。
Objective Comparative studies on the influence of human adult pancreas of different blood group used as antigen substrate upon the specificity and sensitivity of ICA assay with immunohistochemical method.Methods The serum specimens of 24 cases of type 1 diabetic patients with positive ICA testing and 23 cases of normal subjects with negative ICA testing were assayed for the ICA with immunohistochemical method using human pancreas of different A,B,O and AB blood group as the antigen substrate.Results Using human pancreas of O blood group as control, three cases of pancreases of A blood group and B blood group as well as two cases of pancreases of AB blood group were used as antigen substrate for assaying the ICA. The results showed that the specificity and sensitivity of ICA assay of 24 ICA positive sera were basically consisent with that of 23 ICA negative sera, while the false positive or false negative result was not seen.Conclusion The results of this paper demonstrated that the human pancreas of A, B or AB blood group may be used as the pancreas of O blood group for assaying ICA with immunohistochemical method, and human pancreas of different blood group as the antigen substrate for assaying ICA would not influence the specificity and sensitivity of the assay.
