首参颗粒对动脉粥样硬化大鼠血管病理及氧化应激的影响 被引量:6

Effect of "Shoushen Granule" on Vascular Pathology and Oxidative Stress in Rats with Atherosclerosis

摘要 目的:观察补肾中药复方首参颗粒对动脉粥样硬化模型大鼠血管病理及氧化应激的影响。方法:将36只SD大鼠随机分为正常组、模型组、中药组和西药组,每组9只。除正常组外,其余各组通过一次性腹腔注射维生素D3及连续饲喂高脂饮食的方法建立动脉粥样硬化大鼠模型。在造模的同时,中药组和西药组分别予灌胃首参颗粒、普伐他汀,共干预12周。实验结束,HE染色观察大鼠腹主动脉病理变化;酶联免疫吸附法(ELISA)检测大鼠血清髓过氧化物酶(MPO)、氧化低密度脂蛋白(oxLDL)、超敏C反应蛋白(hs-CRP)的变化。结果:病理检测显示,模型组有明显的动脉粥样硬化病变,而药物干预组血管病变明显减轻或消失;模型组MPO、oxLDL、hs-CRP含量较正常组显著升高(P<0.05或P<0.01),药物干预组MPO、oxLDL、hs-CRP含量较模型组显著降低(P<0.01),且中药组和西药组间无明显差异。结论:首参颗粒对动脉粥样硬化病变有明显的防治作用,其机制可能与调节氧化与炎症相关指标有关。 Objective: To observe the effect of "Shoushen Granule" on vascular pathology and oxidative stress in rats with atherosclerosis. Methods: Thirty-six rats were randomized into 4 groups : normal group, model group, Chinese medicine group and western medicine gToup, 9 rats in each group. Except normal group, atherosclerotic rat models were established by high-fat diet and vitamin D injection. At the same time, the Chinese medicine group was treated with "Shoushen Granule" and western medi- cine group was treated with pravastatin for 12 weeks. The abdominal aorta was observed by HE staining and the MPO, oxLDL and hs-CRP were detected by ELISA. Results: Pathological examination showed that atherosclerosis could be found in model group and the vascular dysfunction was relieved or disappeared in medicine groups ; the MPO, oxLDL and hs-CRP were higher in model group than in normal group (P 〈0.05 or P 〈0.01 ), the MPO, oxLDL and hs-CRP were lower in medicine groups than in model group (P 〈0. 01 ), no significant difference was found between Chinese medicine group and western medicine group. Conclusion: "Shoushen Granule" has positive effect on atherosclerosis by adjusting oxidation and inflammation indexes.
